Isolation of a group-specific polysaccharide from tissues infected with Mycobacterium leprae.

چکیده
Investigation of the immunologically active substance in Mycobacterium Zeprae has been hampered because of the unavail ability of pure cultures. However, this difficulty can now be circumvented in part by taking advantage of the identification of the group-specific antigen, Polysaccharide I (PolyINb ), present in Nocardia and Mycobacteria ( I. 3 G. 8) . This has been chemically characterized as a polymer of D-arabofuranose and D-galactopyranose in a molar ratio 3:1 (4) . The present paper deals with the identification of the group-specific polysaccharide (PolyINb ) in extracts of human leproma, after physical removal and enzymatic destruction of all other components that might have been immunologically active.

A RAPID AND SIMPLE METHOD OF DNA EXTRACTION FROM CLINICAL SPECIMENS CONTAINING MYCOBACTERIUM LEPRAE FOR PCR ASSAYS
We describe a rapid, simple, and reliable procedure for routine isolation of Mycobacterium leprae DNA from slit-skin swab specimens. This one-step DNA extraction method is based on the utilization of ChelexR 100, a chelating ion exchange resin. Slit-skin swab specimens from seven untreated leprosy patients at Baba Baghi Leprosy Hospital in Iran were processed by this procedure. The polymer...

Evidence has accumulated in recent years for the presence of common antigens in Mljcobacterium tuberculosis and Mljcohacteriunt leprae. Thus, Pepys et al. (9) , using precipitin tests found thrcc antigens common to the various PPD preparations and lepromin . Tuma and Silva ( IU) observed positive precipitins when antigens isolated from M. tuberculosis and BCG reacted with sera from rabbits immu...
متن کاملEnumeration of Mycobacterium leprae Using Real-Time PCR
Mycobacterium leprae is not cultivable in axenic media, and direct microscopic enumeration of the bacilli is complex, labor intensive, and suffers from limited sensitivity and specificity. We have developed a real-time PCR assay for quantifying M. leprae DNA in biological samples. Primers were identified to amplify a shared region of the multicopy repeat sequence (RLEP) specific to M. leprae an...
متن کاملMolecular Assays for Determining Mycobacterium leprae Viability in Tissues of Experimentally Infected Mice
BACKGROUND The inability of Mycobacterium leprae to grow on axenic media has necessitated specialized techniques in order to determine viability of this organism. The purpose of this study was to develop a simple and sensitive molecular assay for determining M. leprae viability directly from infected tissues. METHODOLOGY/PRINCIPLE FINDINGS Two M. leprae-specific quantitative reverse transcrip...
متن کاملDetection of Mycobacterium avium subsp. paratuberculosis in the mesenteric lymph nodes of goats by PCR and culture
The efficacy of bacterial cultures and IS900-specific polymerase chain reaction (PCR) was compared for the detection of Mycobacterium avium subsp. paratuberculosis (MAP) from the mesenteric lymph nodes of goats. Samples were collected from 75 goats slaughtered in Ilam, in southwest of Iran. Tissue homogenates were inoculated onto four media. The genomic DNA was extracted directly from mesenteri...
